Posted on 2008-10-18 05:16:45

EntreMed. Inc. (Nasdaq: ENMD) a clinical-stage pharmaceutical company developing therapeutics for the treatment of cancer and inflammatory diseases announced that it has commenced a multi-center Phase 2 clinical trial with MKC-1 in patients with advanced pancreatic cancer. The primary objectives of this study will be to determine the antitumor activity of orally-administered MKC-1 in unresectable or metastatic pancreatic cancer patients who have failed at least one prior chemotherapy regimen. The study will also assess the safety tolerability and overall median survival time of pancreatic cancer patients treated with MKC-1. Massachusetts General Hospital Cancer Center is the lead institution for the study and Eunice Kwak. M. D.. Ph. D.. Assistant in Medicine. Tucker Gosnell Center for Gastrointestinal Cancers will serve as the principal investigator. MKC-1 is a novel orally-active cell cycle inhibitor with in vitro and in vivo efficacy against a broad range of human solid tumor cell lines including multi-drug resistant cell lines. Prior preclinical studies have demonstrated that MKC-1 has significant antitumor activity as both a single agent and in combination with an approved epidermal growth factor/epidermal growth factor receptor (EGF/EGFR) inhibitor in pancreatic cancer models. MKC-1 has also demonstrated broad-acting antitumor effects including tumor growth inhibition or regression in multiple preclinical models. MKC-1 has been shown to inhibit mitotic spindle formation prevent chromosome segregation in the M-phase (mitosis) of the cell cycle and induce apoptosis. Carolyn F. Sidor. M. D.. M. B. A.. EntreMed Vice President and Chief Medical Officer commented on the study. “Pancreatic cancer patients continue to need more effective and better tolerated treatment options. MKC-1 has shown good activity in preclinical testing against pancreatic tumors supporting its use in this patient population. Positive results from this clinical trial will provide the basis for designing future randomized Phase 2 studies. With this clinical trial initiation we are now evaluating MKC-1 as a single agent or in combination in patients with pancreatic cancer metastatic breast cancer non- small cell lung cancer (NSCLC) and leukemia.” Pancreatic cancer is the abnormal cell growth in the tissue of the pancreas. The pancreas is an organ about six inches in length which is located behind the stomach next to the small intestine. The pancreas produces enzymes that aide in the digestion and absorption of food. Unless pancreatic cancer is detected early it is difficult to control. At the present time there is no effective screening for pancreatic cancer and because the pancreas is hidden by other organs it is difficult to diagnosis. Pancreatic cancer is the fourth leading cause of cancer death in the United States. The American Cancer Society estimates that approximately 37,000 Americans will be diagnosed with cancer of the pancreas in 2007 resulting in approximately 33,000 deaths. EntreMed. Inc is a clinical-stage pharmaceutical company developing therapeutic candidates primarily for the treatment of cancer and inflammation. Panzem(R) NCD (2-methoxyestradiol or 2ME2) is currently in multiple Phase 2 clinical trials for cancer. MKC-1 an oral cell-cycle regulator is in multiple Phase 1 and 2 studies for cancer. ENMD-1198 a novel tubulin-binding agent is in Phase 1 studies in advanced cancers. Panzem(R) is also in preclinical development for rheumatoid arthritis and ENMD-2076 a dual-acting Aurora-angiogenesis inhibitor is in preclinical development for cancer. It is hard to talk about but even harder to talk about finances to everyone here but I need to know if anyone else has had this problem..... bad news is my mother does not undergo insurance. Her and my daddy were self employed for years and when it came measure to paying the insurance premiums after they retired they had to end - eat or undergo insurance. Daddy is ok he is 69 and has Medicare. Mom is too young. 62 1/2 (funny how I add in the 1/2 now desire we did when we were kids)   We are reaching out to the hospital for there program. Also we have been told that Social Security Diability is something she needs to apply for. Since it is a terminal illness can she receive medicare early instead of waiting until she is 65. (which we experience she will not 2 1/2 years seems to be a desire time right now) Has anyone done this?   Also any other ideas you could share with myself and my family on anything related to this disease would be greatly appreciated. Lastly the cancer care treatment center is requesting $500 drink and a promisory note to be singed tomorrow before she can acquire her chemo. "Obesity, Lack Of Exercise May Increase Cancer Risk" posted by ~Ray

Obesity and aversion to exercise may change magnitude the risk of pancreatic cancer inform scientists. Studies have earlier indicated that smoking is responsible for about 25 percent of pancreatic cancer cases. But researchers of the Dana Farber Cancer Institute in Boston examined 144 pancreatic cancer patients and 429 populate without the disease and open that obesity and lack of exercise may be for a similar percentage of cancer patients. Pancreas the gland under the curvature of the stomach produces enzymes that are useful for digestion and secretes hormones that help maintain and adjust be sugar levels. "Pancreatic cancer, the most fatal cancer?" posted by ~Ray
Posted on 2007-11-06 01:17:21

Luciano Pavarotti was buried last weekend. He was diagnosed with pancreatic cancer about a year ago and until lately seemed to be doing come up. I actually assumed the diagnosis was an error because he lived so long. This year about 37,000 populate will be diagnosed with this cancer and 33,000 ordain die of it. The 5-year survival from this disease is around 5% and only a quarter of patients live past the first year. The only treatment is surgery and this is rarely successful in curing populate. Only 7% of people with this disease are found at an early re-create and of these only one-fifth is cured with surgery. Chemotherapy can help a little and there are newer drugs that may add to the benefits of chemotherapy but not much. At beat all this treatment buys a bring together of months more or less. comfort populate want treatment. Many years ago the only medicate available was 5-fluorouracil which was never proven to help. At that time I saw a psychiatrist (he was the patient) who had been recently diagnosed with advanced pancreatic cancer. My advice was to avoid chemotherapy-why make yourself feel worse with no up align. But he couldn’t deal with no treatment and went to another oncologist to get chemotherapy. The medicate we now use to treat advanced pancreatic cancer gemcitabine does help but as I said only a little. Many people have had some temporary relief of their symptoms mainly pain with this drug. It is usually given along with one of many other chemotherapy drugs but there hasn’t been any evidence that adding another drug is better than gemcitabine alone. Another problem with pancreatic cancer is that it is nearly impossible to detect early. There is no screening test that will sight early pancreatic cancers. This is why it is rare to find someone with just a small tumor that hasn’t move. The only instances where it is found early is when it blocks the bile duct and causes a backup of bile. This turns the person color and is easy to sight. But this is rare. In fact there are a few people with a hereditary form of the disease and the strategy there is just to direct to shift the pancreas before there is a chance for cancer to develop. Heredity is a small risk factor for pancreatic cancer. The major ones are smoking and the one that sealed Pavarotti’s ordain obesity. Of course his extra weight may undergo kept him alive a bit longer than I would have expected. Not a great strategy. So as I keep saying eating healthy is still the beat strategy for avoiding many cancers.
